Makarios Griniezakes was born in the city of Heraklion, in Crete. he graduated from the Higher Ecclesiastical School of Athens, and the School of Theology of the Kapodistrian University of Athens. He pursued postgraduate studies in Christian Ethics at the School of Theology of Boston University, in History of Science at the Faculty of Arts and Sciences of Harvard University, as well as in Bioethics at the Faculty of Science of Monash University. He received his doctoral degree from the Faculty of Medicine of the University of Crete, presenting his dissertation under the title of Κλωνοποίηση: Ἠθικοκοινωνικές καί Θεολογικές Συνιστῶσες (Cloning: Moral-social and theological aspects). Since 2009 he has been member of the Pan-Orthodox Committee of Bioethics. In 2008 he was nominated by the Ecumenical Patriarch Bartholomew as Archimandrite of the Ecumenical Throne at the Phanar. From 2005 to 2014 he was member of the Synodal Committee of the Church of Crete on Bioethics and Contemporary Issues. He is lecturer at the Patriarchal University Ecclesiastical Academy of Crete. In 2015 he was nominated as auxiliary bishop of Christoupolis.
